Ovation® FFPE WTA System

NuGEN's latest product for gene expression profiling from FFPE samples

The NuGEN Ovation FFPE WTA System (Part No. 3403) is a whole transcriptome amplification solution that enables gene expression studies using precious archives of small and degraded RNA in FFPE samples. The system is comprised of reagents and a protocol required for amplification of as little as 50 ng of FFPE-derived total RNA. The protocol takes just five hours, and can be used for qPCR, sample archiving, fragmentation, and labeling.

NuGEN's latest generation FFPE solution, the Ovation FFPE WTA System, allows researchers to obtain even more sensitivity and reproducibility with a simplified workflow. The new system offers increased %P values to detect more transcripts, lower Actin/GAPDH ratios for improved coverage and higher reproducibility of technical replicates for superior array performance. Additional enhancements to this new product include single-step amplification and an overall protocol length shortened by one hour to even further streamline your workflow.

The amplified cDNA can be fragmented and labeled in less than two hours for analysis on Affymetrix GeneChip® 3', Exon ST, and Gene ST expression arrays using NuGEN's Encore™ Biotin Module.

The Ovation FFPE WTA System provides optimized reagent mixes and a protocol to process 12 or 60 RNA samples (Part No. 3403-12, 3403-60) and is also available in a 96 reaction size for automation workflows (3403-A01).

NOTE: With the release of Ovation FFPE WTA System, WT-Ovation FFPE System V2 (Part No. 3400-12) has been discontinued.

Array Performance Metrics with FFPE RNA: cDNA targets were prepared from triplicate determinations of RNA extracted from formalin fixed paraffin embedded tissue (FFPE) using either Ovation FFPE WTA System or WT-Ovation FFPE System V2. The amplified cDNA was fragmented and labeled using the Encore Biotin Module, and hybridized to GeneChip® Human Genome U133A 2.0 arrays. The data shows an increase in %P values with the Ovation FFPE WTA Systems together with reduced Actin and GAPDH ratios which illustrates improvements in both transcript coverage and representation of less abundant transcripts.
